Marisa Paredes began studying music and drama when she was 14 years old. She had her theatrical debut under the direction of José Osuna and in 1960, when she was 14, she had a part in José María Forqué's television crime/drama 091, Police Speaking (091, Policía al habla). She started working in cinema during the 1960s in films like Gritos en la noche (1962) by Jesús Franco, El mundo sigue (1965) by Fernando Fernán Gómez and Las salvajes en Puente San Gil (1966) by Antoni Ribas.

Her film career took off thanks to Pedro Almodóvar who asked her to participate in High Heels (Tacones lejanos, 1991), along with Victoria Abril and Miguel Bosé. Her role as Becky del Páramo earned her a Spanish Actors Union Film Award and she became known as "an Almodóvar girl." Four years later she starred in the director's The Flower of My Secret (La flor de mi secreto), for which she received a Goya nomination.

The actress has worked with filmmakers like Raúl Ruiz Three Lives and Only One Death (Tres vidas y una sola muerte, 1996), Arturo Ripstein Deep Crimson(Profundo carmesí, 1996) and No One Writes to the Coronel (El coronel no tiene quien le escriba, 1999), Roberto Benigni Life is Beautiful (La vida es bella, 1997) and Manuel de Oliveira Magic Mirror (Espelho Mágico, 2005). Paredes served as president of the Spanish Movie Academy from 2000 to 2003. The Skin I Live In (La piel que habito), her most recent film, was also directed by Almodóvar. The film premiered in competition at the Cannes Film Festival in 2011.
